The First Regenerable Streptavidin Biosensor

Break free from costly, one-sample-per-sensor workflows with the Gator® Regen SA (RSA) Biosensor. Regenerate, reuse, and reduce cost without compromising performance.

Streptavidin biosensors have always been one-and-done. Load your biotinylated target, run the interaction, discard the probe. Regen SA (RSA) probes change that. Built on an engineered streptavidin, RSA regenerates with the same standard protocol you’d use on any reusable capture surface, so a single probe measures up to 10 interactions. Paired with Gator Bio’s XT optics, RSA delivers up to 4x higher signal at up to 8x lower cost per interaction than non-reusable SA sensors.

Consistent recapture across a wide dynamic range

RSA probes demonstrate robust regeneration performance using the proprietary Regeneration Solution for RSA which effectively removes both the bound ligand and analyte while leaving the streptavidin layer intact. Consistent loading height with recovery of 90-110% is achieved across 10 recapture cycles.

10 recapture cycles of hIgG labeled with biotin-LC-LC on RSA probes at concentrations of 0.625 – 80 μg/mL demonstrating 1.3-5.8% CV on signal loading with 90-110% recovery across all concentrations.

Kinetic characterization of biotinylated CRP and PD-L1 with their respective antibodies on the same set of RSA probes with alternating recapture cycles.

Capture and screen multiple biotinylated samples with a single reusable biosensor

Move through your screening libraries more efficiently by capturing up to 10 different biotinylated samples on the same probe with reliable results.  Maintain consistent kinetic performance by using same biosensor for multiple samples.

Up to 4x higher signal enabled by XT optics technology

XT optics technology increases the signal per bound molecule, improving total signal up to 4x and allowing users to use load fewer ligand molecules to get the same amount of signal as traditional BLI biosensors. This allows users to limit mass transport and avidity effects with ease.

Comparison of signal height for CRP/antiCRP kinetics interaction on (A) SA probes and (B) RSA probes.

“The Regen SA probes have been an absolute game-changer for our kinetics screening workflows. What we love most is the streamlined, simple workflow; it operates exactly like any other reusable capture assay, allowing us to increase our sample throughput and save us valuable time. We’ve been so impressed that we plan to adopt Regen SA as our primary probe for biotin-tag capture assays.”

Enhanced sensitivity and dynamic range for mouse antibody screening

Gator MFC XT Biosensors are next generation anti-mouse IgG Fc capture sensors engineered for superior performance and broader dynamic range. They support both quantitation and kinetic analysis of mouse IgG antibodies and mouse Fc-fusion proteins with significantly improved sensitivity over previous MFC biosensors.

The MFC XT sensors exhibit high affinity for all four mouse IgG subclasses (IgG1, IgG2a, IgG2b, IgG3) with minimal cross-reactivity to rat, human, and bovine IgG. They are suitable for measuring mouse IgG in both crude and purified samples. With up to a 4-fold increase in ligand binding capacity, the sensors provide enhanced kinetic characterization of small proteins and enable detection of low-abundance mouse antibodies in hybridoma cultures.

MFC XT Biosensors can be regenerated up to 20 times with minimal loss of performance, delivering a cost-effective solution for high-throughput applications such as lead identification, hybridoma and cell line development, process optimization, and QC testing

Key Features

MFC XT’s higher signal provides better separation, even at high concentrations

The MFC XT biosensors deliver significantly higher binding signals compared to competitor sensors, resulting in greater curve separation across a wide concentration range. This improved resolution is especially important at higher concentrations, where competitor data compresses and overlaps, making it difficult to distinguish between samples. With stronger signal and broader dynamic range, MFC XT enables more accurate quantitation and kinetic analysis, even in challenging assay conditions.
